Reconstitution Guide

Reconstitution Guide (with Dilution Examples)

This guide explains how lyophilized peptides are typically reconstituted using bacteriostatic water (BAC water). It is intended for informational and laboratory handling purposes only. Always follow strict sterile technique and seek professional guidance where appropriate.

What is Reconstitution?

Reconstitution is the process of adding a sterile diluent (commonly bacteriostatic water) to a freeze-dried peptide vial in order to form a liquid solution suitable for handling and further preparation.

What You Need

  • Lyophilized peptide vial (10mg / 20mg / 30mg / 40mg)
  • Bacteriostatic water (BAC water)
  • Alcohol swabs (70% isopropyl alcohol)
  • Sterile syringe and needles
  • Clean, sterile workspace

General Principle

Once BAC water is added, the concentration depends on the amount of liquid used.

Formula:

Concentration (mg/mL) = Total peptide (mg) ÷ Total BAC water added (mL)

Example Dilution Chart (BAC Water Reconstitution)

10mg Vial

BAC Water AddedFinal Concentration
1 mL10 mg/mL
2 mL5 mg/mL
3 mL3.33 mg/mL
4 mL2.5 mg/mL

20mg Vial

BAC Water AddedFinal Concentration
1 mL20 mg/mL
2 mL10 mg/mL
3 mL6.67 mg/mL
4 mL5 mg/mL

30mg Vial

BAC Water AddedFinal Concentration
1 mL30 mg/mL
2 mL15 mg/mL
3 mL10 mg/mL
4 mL7.5 mg/mL

40mg Vial

BAC Water AddedFinal Concentration
1 mL40 mg/mL
2 mL20 mg/mL
3 mL13.33 mg/mL
4 mL10 mg/mL

Important Handling Notes

  • Always inject BAC water slowly down the side of the vial
  • Do not shake vigorously — gently swirl instead
  • Allow powder to fully dissolve before use
  • Use sterile equipment at all times
  • Discard if solution appears cloudy or contaminated

Storage After Reconstitution

  • Store at 2–8°C (refrigerated)
  • Do not freeze
  • Keep vial sealed and protected from light
  • Use sterile technique each time the vial is accessed
